Experimental Studies On The Effect Of Tetracycline Administration On Tooth Development


Abstract
The author has studied the effect of tetracycline antibiotics on tooth development of rabbits weighing about 2 kg by light and fluorescent microscopy.
A. It were used intravenous tetracycline HCl and intramuscular oxytetracycline as experimental antibiotics, and 1% liquid alizarin red S 10 c.c. as fluorescent indicative liquid.
B. 7 groups of experiment including 21 animals were carried out, namely:
1. Animals were injected i.m. with a dose of 30mg/kg body weight of oxytetracycline for 5 days from 1st- to 6th day of experiment, and i.v. of 1.5 mg/kg weight of tetracycline HC1 on 11th day.
2. Animals were injected i.v. with a dose of 1.5mg/kg weight tetracycline HCI on 1st day of experiment, and i.m. of 50 mg/kg weight of oxytetracycline for 5 days from 6th- to 11th day.
3. Animals were injected i.v. with a dose of 1.5 mg/kg weight of tetracycline HCl on 1st day of experiment, and i.m. with one of 20 mg/kg weight of oxytetracycline for 5 days on 6th- to 11th day.
4. Animals were injected i.m. of alizarin red S on 1st day of experiment, and with a dose of 20mg/kg weight of oxytetracycline for 5 days from 6th- to 11th day.
5. Animals were injected i.m. with alizarin red S on 1st day of experiment, and i.v. with a dose of 5mg/kg weight of tetracycline HCI for 5 days from 6th- to 11th day.
6. Animals were injected i.m. with alizarin red S on 1st day of experiment, and i.v. with one of 2 mg/kg weight of tetracycline HCl for 5 days from 6th- to 11th day.
7. Control groups of animal were injected i.v. with a dose of 1. 5mg/kg weight of tetracycline HC1 on
1st day of experiment, and i.m. with one of 20mg/kg weight of oxytetracycline on 5th- and 11th day.
All animals were decapitated by air injection on 13th day, the termination of experiment.
C. Observation method: Upper and lower jaw including incisors & molars removed from rabbits were fixed 10% formalin, deivided incisors & molars, and made the ground sections. Author observed the deposition of tetracycline antibiotics on enamel, dentin, and cerrentum of tooth, and the degree of fluorescent brightness & fundamental formation according to the injected method & dose of antibiotics by AO spenser fluorescent- and Nikon light microscopy.
D. The following results were obtained:
1. In the dentine layer, the indicative lines of interference with calcification of the matrix corresponding to the serial injection of tetracycline appeared more prominent by intramuscular injection than by intravenous injection, and by over-dose than by small dose. The distance between the infered calcified lines was gradually decreased by serial injection.
2. The tetracycline deposited line exhibited a golden yellow fluorescent one under the ultraviolet light.
3. The width of fluorescent line and the brightness of fluorescence were in proportion to the persistence of tetracycline in tissue, and to the number of administration in serial injection.
4. The dentin formation was remarkably decreased by the serial injection of tetracycline.
5. The deposition of the tetracycline complex were the same regardless the dose and the method of injection.
